Angiopoietin-like protein 5 (ANGPTL5)

Angiopoietin-like protein 5 (ANGPTL5) is a secreted glycoprotein and member of the angiopoietin-like protein family, sharing structural features with angiopoietins such as an N-terminal coiled‑coil domain and a C-terminal fibrinogen-like domain[1]. ANGPTL5 is predominantly expressed in adipose tissue, with lower levels in heart, ovary, testis, skin, bronchial, and epididymal tissue, and plays a regulatory role in lipid and triglyceride metabolism through inhibition of lipoprotein lipase (LPL)[2]. Elevated circulating ANGPTL5 is associated with obesity, diabetes, and cardiovascular disease, and rare loss-of-function mutations have been linked to lower plasma triglycerides and reduced BMI, suggesting its importance in metabolic homeostasis[2]. Additionally, ANGPTL5 affects hematopoietic stem cell proliferation and engraftment and may be involved in inflammatory processes, lipid oxidation, and stem cell biology[1][2][3]. There are no currently approved drugs targeting ANGPTL5 directly, but modulation of related ANGPTL proteins and mechanisms is an area of therapeutic research[2]. ANGPTL5’s function appears non-redundant with other family members and its biology is still incompletely characterized, as no mouse ortholog exists[2][3].
