Overview

The interaction between angiotensin-converting enzyme 2 (ACE2) and the spike glycoprotein S1 subunit is a critical molecular event that enables the entry of coronaviruses, such as SARS-CoV-2, into host cells. The spike (S) protein of the virus contains two main subunits: S1, which mediates receptor binding, and S2, which facilitates membrane fusion. The receptor-binding domain (RBD) within the S1 subunit specifically recognizes and binds to ACE2 on host cell surfaces. Disruption or inhibition of this interface blocks infection—making it a prime target for antiviral drugs and neutralizing antibodies.
