Overview

ANKDD1A is a protein-coding gene located at chromosome 15q22.31 encoding a protein with nine ankyrin repeats and a death domain. Ankyrin repeats are motifs that mediate protein–protein interactions, while the death domain is associated with apoptosis and inflammatory signaling. ANKDD1A acts as a tumor suppressor in glioblastoma, primarily under hypoxic conditions. It directly interacts with factor inhibiting HIF1α (FIH1), regulating the transcriptional activity and stability of HIF1α, an important factor in cellular adaptation to hypoxia. By upregulating FIH1, ANKDD1A decreases glucose uptake, lactate production, inhibits autophagy, and induces apoptosis in GBM cells. ANKDD1A is frequently hypermethylated in gliomas, suggesting its role as a tumor-specific epigenetic biomarker and potential therapeutic target[1][4][6][7][9][11][12].
