Overview

Ankyrin repeat and kinase domain containing protein 1 (ANKK1) is a member of the serine/threonine protein kinase family, with structural domains including an N-terminal kinase domain and C-terminal ankyrin repeats[4][3][1]. These features enable it to phosphorylate substrates and participate in protein–protein interactions within signal transduction pathways. ANKK1 is located adjacent to the dopamine D2 receptor gene (DRD2), and genetic variants in ANKK1 (notably Taq1A) are associated with altered dopamine receptor density and a spectrum of neuropsychiatric and addiction phenotypes[1][3][2][5]. Functionally, ANKK1 is implicated in neurogenesis, neuronal differentiation, cell migration, and regulation of dopaminergic and NF-κB signaling pathways[1][2][3]. The protein exists in multiple isoforms, and is expressed in both central nervous system and muscle precursor cells, where it may contribute to development and cellular signaling[1].
