Ankyrin repeat and SOCS box protein 1 (ASB1) is a protein encoded by the ASB1 gene in humans, characterized by the presence of six N-terminal ankyrin repeats and a C-terminal SOCS box domain. ASB1 is a member of the SOCS (Suppressor of Cytokine Signaling) box protein family, which typically function as substrate-recognition components of multimeric E3 ubiquitin ligase complexes, such as the Elongin-Cullin-SOCS box (ECS) complex. Unlike many other SOCS box proteins, ASB1 does not promote ubiquitin-mediated proteasomal degradation of its primary binding substrate (TAB2); instead, it stabilizes TAB2 by inhibiting K48-linked polyubiquitination, enhancing NF-κB and MAPK pathway signaling and promoting pro-inflammatory responses. ASB1 has been associated with several human diseases, including immune dysfunction, cardiovascular conditions, and neurodevelopmental disorders, and it may serve as a genetic/epigenetic marker in those contexts. However, there is no evidence that ASB1 is directly targeted by drugs or established as a therapeutic target.
