Overview

Ankyrin repeat and SOCS box protein 10 (ASB10) is a member of the ankyrin repeat and SOCS box-containing (ASB) family of proteins and functions as an E3 ubiquitin ligase, regulating protein stability and degradation through ubiquitin-mediated processes[1][2][3]. ASB10 is expressed abundantly in ocular tissues like the trabecular meshwork, where it modulates intraocular pressure and has been genetically linked to primary open-angle glaucoma[1][2]. It exerts its function by coupling ankyrin repeat-mediated substrate recognition with the SOCS box, which recruits ubiquitination machinery, targeting proteins for degradation by the proteasome or autophagy-lysosome pathway[1][2][4]. In cardiac tissues, ASB10 interacts with and stabilizes HSP70, exacerbating pathological cardiac remodeling under stress[3]. In cancer, ASB10 mediates degradation of TEM8 in response to estrogen receptor-α in triple-negative breast cancer, impacting cancer stem cell properties and neovasculogenesis[1]. Thus, ASB10 is an important regulator of protein quality control with roles in ocular physiology, cardiovascular disease, and cancer biology.
