Ankyrin repeat and SOCS box protein 8 (ASB8) is a member of the ankyrin SOCS box (ASB) protein family characterized by four ankyrin repeats and one SOCS box domain. The protein functions primarily as a substrate-recognition component in SCF-like ECS (Elongin-Cullin-SOCS-box) E3 ubiquitin-protein ligase complexes, facilitating the polyubiquitination and subsequent proteasomal degradation of specific target proteins. Through this process, ASB8 regulates protein turnover, intracellular signaling pathways—including the inhibition of IFN-beta production by targeting TBK1 for degradation—and may influence cell proliferation and cancer development, notably in lung carcinoma. ASB8 exhibits highest expression in skeletal muscle, with additional expression in heart, brain, placenta, liver, kidney, and pancreas. Although classified within the E3 ubiquitin ligase complex components, it is not currently a direct drug target nor do specific drugs or biomarkers linked to ASB8 exist, though its modulation could have therapeutic relevance in oncology and immunology.
