Ankyrin repeat domain-containing protein 1 (ANKRD1)

Ankyrin repeat domain-containing protein 1 (ANKRD1 or Cardiac ankyrin repeat protein/CARP) is a cardiac- and skeletal muscle-enriched protein belonging to the muscle ankyrin repeat protein family[1][3][6]. It contains five tandem ankyrin repeats and plays key roles in transcriptional regulation, assembly of the sarcomere (the basic contractile unit of muscle), and mechanosensing of cardiac stress signals[1][3][6]. ANKRD1 acts as a nuclear transcription factor that negatively regulates cardiac gene expression, and is highly induced by cardiac stress and in various cardiomyopathies and heart failure states[1][3][5][6]. Its levels and activity are implicated in the pathogenesis of several heart and muscle diseases, and it is being explored as a sensitive and specific biomarker for certain cardiac and muscle diseases, as well as for predicting chemotherapy outcomes in ovarian cancer[3]. No direct modulating drugs are currently listed, and mechanisms of action for drug-target interactions are not established in available literature.
