Ankyrin repeat domain-containing protein 12 (ANKRD12) is a large nuclear protein encoded by the ANKRD12 gene and is a member of the ankyrin repeats-containing cofactor family[1][3]. ANKRD12 contains N-terminal ankyrin repeats and a C-terminal co-activator-interacting domain and functions as a versatile transcriptional coregulator, generally acting as a corepressor by recruiting histone deacetylases (HDACs) and interacting with transcription coactivator complexes such as p160 and ADA3[1][2][3]. It can modulate nuclear receptor–mediated transcription, and alternative splicing produces multiple isoforms. ANKRD12 has been shown to be downregulated in colorectal cancer tissues, and lower expression is associated with higher frequency of liver metastasis and poor overall survival in patients, identifying it as a potential prognostic marker in cancer[1]. Its mechanism involves inhibiting ADA3-mediated transcriptional co-activation on nuclear receptors, linking it to regulation of gene expression and cancer pathogenesis[1][2]. Currently, there are no known drugs that directly target ANKRD12, nor are there documented therapeutic safety concerns.
