Overview

Ankyrin repeat domain-containing protein 13D (ANKRD13D) is a member of the ankyrin repeat domain (ANKRD) 13 family that contains ubiquitin-interacting motifs[2][12]. This integral membrane-associated protein specifically recognizes and binds Lys-63-linked ubiquitin chains, distinguishing them from Lys-48 chains, and regulates the internalization of ligand-activated receptors such as EGFR by binding to their ubiquitinated forms at the cell membrane[2][3][13]. ANKRD13D’s function is implicated in receptor endocytosis and trafficking, acting as a molecular scaffold and negative regulator of receptor internalization[1][5][6][11]. ANKRD13D is reported as a potential tumor-specific target in renal cell carcinoma and may act as an immune regulator, opening avenues for immunotherapeutic approaches in oncology[4]. There are no known drugs approved to target ANKRD13D, no established mechanisms of drug action, nor established biomarker or safety concern profiles[2][4][12].
