Ankyrin repeat domain-containing protein 2 (ANKRD2, also called ARPP) is a member of the muscle ankyrin repeat protein (MARP) family, highly expressed in cardiac and skeletal muscle. It contains ankyrin repeats, a PEST-like and lysine-rich N-terminal region, and a proline-rich C-terminal region. ANKRD2 is primarily associated with titin at the sarcomere I-band and translocates to the nucleus under stress, linking mechanical muscle signals to gene regulation. It interacts with transcription factors (p53, PML, NF-κB p50, YB-1) and signaling proteins (Akt2), modulating muscle differentiation, repair, and inflammatory signaling. Its expression is induced following muscle injury and altered in several pathological conditions (cardiac, neurodegenerative, muscular, and cancer). While not a traditional drug target, its modulation may impact muscle physiology and disease progression
