Ankyrin repeat domain-containing protein 24 (ANKRD24) is a large intracellular protein characterized by multiple ankyrin repeat domains, which are known for mediating protein-protein interactions[1][7][8]. ANKRD24 is primarily expressed in the inner ear, where it is localized to the rootlets of hair cell stereocilia and is essential for their structural integrity[2][3][4][5][7]. It physically surrounds and binds TRIOBP-5, a critical actin-bundling protein, thereby anchoring stereocilia to the cuticular plate and helping to protect the hair bundle from overstimulation and noise-induced damage[2][3]. Disruption of ANKRD24 in mice results in progressive hearing loss and reduced recovery from auditory trauma[3]. Beyond the auditory system, altered expression of ANKRD24 has been recognized as part of prognostic gene signatures in neuroblastoma and is upregulated in certain cancers, suggesting a potential biomarker role[2]. Despite its well-defined structural role in the ear, no current evidence supports its classification as a therapeutic target, receptor, enzyme, or transporter; it is best considered a scaffold protein with disease relevance as a biomarker[1][2][3][7][8].
