Ankyrin repeat domain-containing protein 27 (ANKRD27, VARP) is a cytosolic multi-domain protein involved in regulating vesicle trafficking, particularly through its roles as a guanine nucleotide exchange factor (GEF) for Rab21, Rab32, and Rab38 small GTPases[1][2][3][7]. It modulates endocytic recycling, SNARE complex assembly, and vesicle membrane fusion events by interacting with key trafficking proteins such as VAMP7, and is implicated in melanocyte function by modulating the trafficking of melanosomal enzymes and organelles[1][4][7]. ANKRD27 may play roles in neurite outgrowth and glucose transporter (GLUT1) endosome-to-plasma membrane trafficking, and its variants have been associated with rare diseases such as Hermansky-Pudlak syndrome and esophagitis[1][4][7]. There is no evidence that it is presently a direct therapeutic target or that it interacts with drugs or serves as a biomarker, but it remains of interest for basic research into vesicular trafficking pathways.
