Overview

Ankyrin repeat domain-containing protein 31 (ANKRD31) is a scaffold protein characterized by multiple ankyrin repeats that facilitate protein-protein interactions in meiotic cells[1][2][3][4][5]. ANKRD31 plays an essential role in initiating meiotic recombination by anchoring double-strand break-promoting factors, especially REC114, to specific locations on chromosomes, including pseudoautosomal regions of sex chromosomes[1][2][4][5]. Loss or dysfunction of ANKRD31 leads to misregulation of double-strand break formation, defective recombination, male meiotic arrest and infertility, and reduced oocyte reserve in females[1][2][4][5]. ANKRD31 is not considered a canonical therapeutic target such as a receptor, enzyme, or transporter, but instead serves as a critical scaffold for meiotic chromosomal events. There are no known drugs directly targeting ANKRD31, and no established roles in cancer, inflammation, or other complex diseases outside germ cell development.
