Overview

Ankyrin repeat domain-containing protein 36A (ANKRD36) is a human protein encoded by the ANKRD36 gene on chromosome 2q11.2, belonging to the ankyrin repeat domain protein family[1][2][3][4]. It is largely uncharacterized, but is predicted to function in ion channel inhibition and is implicated in a variety of cellular processes, including transcription initiation, cell cycle regulation, cytoskeletal and ion transport, signal transduction, and apoptosis[1][4]. Emerging research indicates roles in chronic myeloid leukemia progression, type 2 diabetes mellitus (inflammation via circANKRD36), chronic inflammation, reproductive function (via sperm maturation networks), and possibly alcohol consumption behavior[1][4]. While there are currently no drugs that directly target ANKRD36, its mutations and expression patterns are under investigation as potential biomarkers for disease progression and patient stratification in cancers (notably CML) and metabolic/inflammatory conditions[1][4]. Interference with ANKRD36 by microRNAs such as miR-144-5p and miR-182 has been shown to impact tumor cell apoptosis and growth in experimental settings, suggesting indirect therapeutic avenues[4]. Its therapeutic targeting remains experimental and largely unexplored.
