Overview

Ankyrin repeat domain-containing protein 49 (ANKRD49) is an evolutionarily conserved protein defined by four ankyrin repeat motifs, which mediate protein–protein interactions essential for diverse cellular processes[1][3][5]. ANKRD49 is highly expressed in mouse and human testes, predominantly localized to the nuclei of germ cells during spermatogenesis, where it modulates autophagy via activation of the NF-κB signaling pathway under conditions like nutrient deprivation[1][5]. In cancer research, ANKRD49 is upregulated in several tumor types, including lung adenocarcinoma and gastric cancer, and has been shown to increase cell invasion and migration through upregulation of matrix metalloproteinases via P38/ATF-2 signaling[2][3]. It also has a reported role in the induction of fetal globin gene (HBG1) expression[5], and is studied as a potential biomarker for cancer prognosis. There is currently no evidence that ANKRD49 serves as a direct therapeutic target or interacts with approved drugs[1][2][5][6].
