Overview

Ankyrin repeat domain-containing protein 53 (ANKRD53) is a protein characterized by the presence of ankyrin repeat motifs, which are structural domains known for mediating protein-protein interactions. ANKRD53 plays a key role in cell division, being required for proper progression through mitosis. It is involved in the alignment of chromosomes at the metaphase plate, the organization of the microtubule cytoskeleton, and the regulation of cytokinesis, the process that physically separates daughter cells at the end of mitosis. ANKRD53 localizes to the spindle pole during cell division and interacts with the protein DDA3 to control spindle microtubule polymerization. Loss of ANKRD53 function leads to defective spindle assembly, chromosome misalignment, delayed mitotic progression, defects in cytokinesis, and genomic instability, which are hallmarks of malignant transformation. In cancer, especially stomach adenocarcinoma (STAD), ANKRD53 expression is downregulated and serves as an independent prognostic biomarker. Additionally, epigenetic modifications, such as promoter hypermethylation, have been observed in some developmental disorders involving chromosomal instability.
