Overview

Ankyrin repeat domain-containing protein 54 (ANKRD54) is an intracellular adaptor protein characterized by four ankyrin repeat domains, with both nuclear localization (NLS) and export signals (NES)[4]. It regulates intracellular signaling, particularly in erythroid (red blood cell) differentiation, and exerts a protein kinase regulator function[1][3]. ANKRD54 is predominantly expressed in the cytoplasm and nucleus[1]. Functionally, it binds with high selectivity to the SH3 domain of Bruton's tyrosine kinase (BTK), facilitating the nucleocytoplasmic shuttling and exclusion of BTK from the nucleus[4]. It also interacts with LYN kinase, although with less affinity than BTK, supporting its role in protein-protein interaction networks implicated in cellular signaling[4]. While ANKRD54's direct disease relevance is not fully delineated, its interactions with kinases involved in hematopoiesis and immune function suggest it may serve as a modulator in these biological processes[4]. Currently, there are no drugs or biomarkers directly based on ANKRD54, nor is it established as a classical druggable target.
