Overview

Ankyrin repeat domain-containing protein 6 (ANKRD6) is a cytosolic scaffolding protein composed of 6 ankyrin repeats, encoded by the ANKRD6 gene (aliases: Diversin, KIAA0957, Diego homolog). It plays a regulatory role in the negative control of canonical Wnt/β-catenin signaling by recruiting casein kinase I epsilon to the β-catenin degradation complex, thereby facilitating phosphorylation and inhibition of β-catenin signaling. ANKRD6 is prominently expressed in the developing mammalian brain and is also involved in planar cell polarity, interacting with core components of the PCP pathway, notably in the inner ear for cellular orientation and sensory function. As the mammalian homolog of Drosophila diego, ANKRD6 is important for developmental processes but is not currently considered a direct receptor, enzyme, or therapeutic target for drug intervention. No disease-associated pathogenic mutations or actionable pharmacological interventions are reported, though variants may associate with certain inherited disorders.
