Overview

Annexin A6 (ANXA6) is a large member (68 kDa) of the annexin protein family, characterized by calcium-dependent binding to phospholipids and a unique structure comprising eight annexin repeats, distinguishing it from other family members with four repeats. It exists in two main isoforms generated by alternative splicing, and is predominantly localized at the plasma membrane and endosomal compartments. Annexin A6 serves as a multifunctional scaffold protein, facilitating membrane trafficking (exocytosis, endocytosis, multivesicular body formation), modulating cholesterol and lipid homeostasis, and organizing protein complexes at specialized membrane domains. It has calcium channel-like activity in artificial membranes and plays a vital role in extracellular matrix mineralization by promoting mineral vesicle formation and nucleation in bone. ANXA6 modulates cytoskeletal interactions and is implicated in the regulation of cell morphology, migration, and signal transduction, with emerging roles in cancer, cardiovascular, and bone pathologies.
