Overview

Annexin A7 (ANXA7) is a calcium-dependent phospholipid-binding protein and a member of the annexin family with diverse functions linked to cell membrane dynamics. Structurally, it contains a unique, highly hydrophobic N-terminal domain and a conserved C-terminal region, supporting roles in membrane fusion, exocytosis, and plasma membrane repair[1][4]. ANXA7 acts in calcium signaling and is required for ESCRT III-mediated shedding and repair of injured membranes[3]. It also regulates endosomal trafficking and growth factor receptor signaling (notably EGFR), with loss or alternative splicing implicated in tumor progression, particularly in brain and prostate cancers[2][3][4]. Its versatile functions also encompass cardiac and skeletal muscle physiology, and roles in several signaling pathways. No targeted therapeutics are currently approved for ANXA7, but its expression patterns and functional loss serve as biomarkers for certain cancers.
