Overview

Anoctamin-8 (ANO8) is a transmembrane protein and member of the TMEM16 (anoctamin) family, characterized as a key tether protein at the junctions between the endoplasmic reticulum (ER) and plasma membrane (PM)[1][2][3]. Unlike canonical members of the anoctamin family that function as calcium-activated chloride channels, ANO8 predominantly supports the assembly of essential calcium signaling complexes—including STIM1, Orai1, SERCA2, IP3 receptors, and PMCA—at PI(4,5)P2-rich compartments on the ER/PM interface[1][2]. ANO8 regulates store-operated calcium entry (SOCE) by controlling STIM1-STIM1 and STIM1-Orai1 interactions, increasing the number and stability of signaling 'puncta' at ER/PM junctions, and facilitating SERCA2-mediated calcium influx into the ER[1][2]. This modulation tightens the control of receptor-evoked calcium oscillations and prevents calcium toxicity, indicating a central role in the fidelity and efficiency of cellular calcium signaling[1][2][3]. ANO8 has eight transmembrane domains and is expressed in multiple tissues, including electrolyte-transporting epithelia such as the trachea, pancreas, colon, salivary glands, prostate, and thyroid gland[3]. Direct drug interactions, clinical targeting, or established disease biomarker roles have not been reported for ANO8[1][2][3].
