Anti-drug antibody formation (ADA formation)

Anti-drug antibody formation refers to the process where the immune system produces antibodies (anti-drug antibodies, or ADAs) in response to the administration of therapeutic protein drugs, mainly biologics like monoclonal antibodies, fusion proteins, and enzymes[2][6][7]. These ADAs can neutralize the therapeutic effects of the drug, alter its pharmacokinetics and bioavailability, and lead to reduced efficacy, loss of treatment response, or adverse immunological reactions such as hypersensitivity[1][4][6]. ADA formation is a major safety and efficacy concern in the development and clinical use of biologic therapies, necessitating routine immunogenicity testing and monitoring during drug development and patient treatment[2][4][8].
