Overview

The anti-histamine-succinyl-glycine (anti-HSG) arm is a specialized antibody binding domain, typically derived from the 679 monoclonal antibody, designed to recognize the synthetic hapten HSG with high affinity (Sharkey et al., 2003). It serves as a critical component in pre-targeted radioimmunotherapy (PRIT) and immunoscintigraphy, most notably within the Dock-and-Lock (DNL) molecular platform (Goldenberg et al., 2012). In this therapeutic framework, a bispecific antibody is first administered to localize to a tumor-associated antigen, such as carcinoembryonic antigen (CEA), while the anti-HSG arm remains vacant. After the bispecific antibody has cleared from the blood, a small HSG-conjugated peptide carrying a diagnostic or therapeutic payload is injected. The anti-HSG arm then captures this peptide at the tumor site, allowing for high tumor-to-background ratios and reduced systemic toxicity compared to directly labeled antibodies (Schoffelen et al., 2014). This system has been extensively studied in clinical trials for various cancers, including colorectal and pancreatic malignancies, using constructs like the TF2 bispecific antibody.
