Overview

The anti-idiotypic antibody 11D10, also known as TriAb, is a murine monoclonal antibody that serves as a therapeutic vaccine by mimicking a specific epitope of the human milk fat globule (HMFG) membrane, specifically the MUC1 protein. In the context of the Jerne idiotypic network, 11D10 is an Ab2 antibody that functions as an internal image of the MUC1 antigen, which is frequently overexpressed and aberrantly glycosylated in adenocarcinomas such as breast, ovarian, and lung cancers. By administering 11D10, the immune system is prompted to generate a specific anti-anti-idiotypic (Ab3) response, consisting of antibodies and T cells that recognize and attack MUC1-positive tumor cells. This strategy is designed to bypass the immune tolerance often associated with tumor-associated antigens by presenting the epitope in a more immunogenic, antibody-based format. Clinical development of 11D10 has focused on its use as a maintenance therapy or in combination with other anti-idiotypic vaccines and chemotherapy to prevent disease recurrence. Notable safety considerations include the development of human anti-mouse antibodies (HAMA) and localized injection site reactions.
