Overview

Anti-idiotypic antibodies that mimic tumor-associated antigen epitopes are engineered to structurally resemble the antigenic determinants of tumor cells. These “internal image” antibodies (specifically Ab2β subtype) can substitute for poorly immunogenic TAAs by serving as surrogate antigens in cancer vaccines. When administered, they stimulate the host’s immune system to generate an immune response—humoral (antibody-mediated) and/or cellular (T-cell)—against the actual tumor antigen, offering a strategy to overcome immune tolerance and target cancer more directly. Racotumomab is a noted example of this technology used as a cancer vaccine. While promising in early studies, anti-idiotypic antibody vaccines have demonstrated safety but only modest clinical efficacy, and are not traditional drug targets but rather a vaccine modality/platform in immuno-oncology. Key clarifying note: This “target” is not a receptor or a discrete protein, but a class of engineered antibodies designed to mimic tumor antigen epitopes and act as active immunotherapeutic agents. The term should not be used as a canonical molecular target in standard target databases.
