Anti-silencing function 1B histone chaperone (ASF1B) is a member of the H3/H4 family of histone chaperone proteins and plays an essential role in chromatin assembly, disassembly, and remodeling during DNA replication, repair, and transcription. ASF1B is a highly conserved protein that interacts with histones H3 and H4, serving as a central hub in the nucleosome assembly pathway and ensuring proper histone supply at sites of nucleosome formation[1][3][5][6][9]. It is particularly important for supporting rapid cell proliferation and is regulated by the tousled-like kinase family (TLK1, TLK2)[7][8]. ASF1B is overexpressed in various cancers, where it promotes cell cycle progression and proliferation, linking its abnormal function to oncogenesis[2]. Unlike its paralog ASF1A (associated with DNA repair and senescence), ASF1B primarily regulates cellular proliferation, and its elevated levels serve as biomarkers in certain cancer types[2][10].
