Overview

Anti-StreptInCor antibodies are protective immunoglobulins, primarily of the IgG class, generated by the host immune system in response to the StreptInCor vaccine candidate. StreptInCor is a synthetic 55-amino acid peptide derived from the conserved C-terminal region of the Streptococcus pyogenes M protein, designed to provide broad-spectrum protection against Group A Streptococcus (GAS) infections. These antibodies function by binding to the conserved C-repeat region of the M protein, which promotes opsonization and subsequent phagocytic clearance of the bacteria while also inhibiting bacterial adhesion to host epithelial cells. A critical advantage of anti-StreptInCor antibodies is their specificity for non-host-reactive epitopes, ensuring they do not cross-react with human cardiac tissues like cardiac myosin, a common complication of other M protein-based vaccines. As such, they are a key focus in the development of a safe, universal vaccine for preventing rheumatic fever and rheumatic heart disease. In clinical development, the measurement of these antibodies serves as a vital biomarker for assessing vaccine-induced immunity and potential efficacy.

Mechanism of action

Anti-StreptInCor antibodies provide protection by binding to the conserved C-terminal region of the M protein on the surface of Streptococcus pyogenes. This binding facilitates opsonization, leading to enhanced phagocytosis and killing of the bacteria by neutrophils and macrophages. Additionally, these antibodies inhibit the adhesion and invasion of the pathogen into host cells, such as pharyngeal epithelial cells, thereby preventing colonization and infection.
