Activation of cytotoxic CD8-positive T lymphocytes occurs when mature dendritic cells process and present intracellular antigens on major histocompatibility complex class I (MHC I) molecules. This presentation is essential for initiating effective CD8 T cell–mediated immunity against viruses, intracellular pathogens, and tumor cells[1][2][3][5][6]. In this process, dendritic cells, especially certain subsets such as CD8α+ cDC1 in mice, are responsible for acquiring, processing (including cross-presentation), and displaying antigenic peptides enabling naive CD8 T cells to recognize antigens and become effector CTLs. The process can be modulated in therapeutic settings (such as cancer immunotherapy) by manipulating dendritic cell or MHC class I function, or using dendritic cell vaccines to promote robust anti-tumor T cell responses[1][2][3][6].
