Overview

Antigen-presenting cell (APC) activation via the MHC class I pathway is a critical process in the adaptive immune response, enabling cytotoxic T lymphocytes (CD8+ T cells) to recognize and eliminate infected or abnormal cells. This pathway involves the presentation of endogenous antigens—typically derived from intracellular pathogens like viruses or from mutated proteins in cancerous cells—on MHC class I molecules at the surface of nearly all nucleated cells. It facilitates immune surveillance and targeted destruction of compromised cells. The pathway involves antigen processing by the proteasome, transport of peptides into the ER by TAP, peptide loading onto MHC-I with the help of chaperones, and surface expression of the peptide-MHC-I complex for T cell recognition.
