Overview

CARGONAUT is a proprietary immune-modulating carrier platform developed by YGION Biomedical, designed to deliver individualized neoantigen peptides to professional antigen-presenting cells (APCs). The platform utilizes a specialized moiety that specifically binds to the CD32 (FcγRII) receptor on APCs, such as plasmacytoid dendritic cells and B cells, to ensure targeted delivery and internalization of the vaccine components. By co-delivering neoepitopes and a molecular adjuvant, such as a TLR9 agonist, CARGONAUT mimics natural viral infection to break immune tolerance and promote the cross-presentation of tumor antigens to the patient's T cells. This approach aims to generate a robust, polyclonal, and tumor-specific immune response while maintaining a high safety profile. The lead candidate utilizing this technology, YG-01, is an individualized cancer vaccine currently in development for the treatment of glioblastoma and other solid tumors.

Mechanism of action

The CARGONAUT carrier is a modular vaccine platform that targets the CD32 (FcγRII) receptor on professional antigen-presenting cells (APCs), such as plasmacytoid dendritic cells (pDCs) and B cells. It delivers tumor-specific neoantigen peptides and a TLR9 agonist (CpG) to these cells, facilitating efficient internalization and cross-presentation of antigens to T cells, thereby triggering a potent and precise anti-tumor immune response.
