Overview

The Antigen-presenting glycoprotein CD1d–alpha-galactosylceramide (CD1d–α-GalCer) complex is a molecular assembly consisting of the MHC class I-like protein CD1d and the synthetic glycolipid agonist alpha-galactosylceramide. CD1d is a non-polymorphic antigen-presenting molecule specialized in displaying lipid antigens to a unique subset of T cells known as invariant Natural Killer T (iNKT) cells (UniProt P15813). When α-GalCer is loaded into the hydrophobic binding pocket of CD1d, the resulting complex is specifically recognized by the semi-invariant T-cell receptor (TCR) of iNKT cells (PubMed: 9311991). This recognition triggers a rapid and robust release of both Th1 and Th2 cytokines, such as interferon-gamma and interleukin-4, which bridge innate and adaptive immunity (PubMed: 10974027). Consequently, the CD1d–α-GalCer complex has been extensively investigated as a therapeutic target for cancer immunotherapy and as a vaccine adjuvant to enhance immune responses (PubMed: 21546910). Drugs targeting this complex, such as KRN7000 and its synthetic analogues, aim to harness the anti-tumor and immunomodulatory activities of NKT cells. However, therapeutic application faces challenges including the potential for iNKT cell anergy and systemic cytokine-related toxicities (PubMed: 24103442).
