The CD1d–glycolipid complex is a molecular assembly consisting of the non-classical MHC class I-like protein CD1d and a lipid-based antigen, which serves as the primary ligand for invariant Natural Killer T (iNKT) cells (UniProt P15813). Unlike classical MHC molecules that present peptide fragments, CD1d features a large, hydrophobic binding groove designed to sequester the hydrocarbon chains of glycolipids, while exposing the sugar headgroup for recognition by the semi-invariant T-cell receptor (TCR) (PubMed: 10359182). This interaction is a cornerstone of the innate-adaptive immune interface, as the formation of this complex leads to the immediate and robust secretion of a broad spectrum of cytokines, including IFN-gamma and IL-4 (PubMed: 21549324). In clinical contexts, the complex is targeted using synthetic agonists like alpha-galactosylceramide (alpha-GalCer) to stimulate anti-tumor immunity or modulate autoimmune responses (PubMed: 24043813). Therapeutic development focuses on optimizing the stability of the CD1d–glycolipid complex to prevent iNKT cell exhaustion and to bias the immune response toward specific cytokine profiles (PubMed: 30108264).
The glycolipid ligand binds within the hydrophobic pockets of the CD1d molecule, presenting the polar head group to the invariant T-cell receptor (TCR) of Natural Killer T (NKT) cells, which triggers rapid cytokine release and immune modulation (PubMed: 17512984).
