Overview

The Antigen-presenting glycoprotein CD1d–invariant Natural Killer T-cell receptor (iNKT TCR) complex is a specialized immunological synapse formed between the MHC class I-like molecule CD1d and the semi-invariant T-cell receptor of iNKT cells. CD1d is a non-polymorphic glycoprotein that presents endogenous and exogenous lipid antigens, such as glycolipids and phospholipids, to the immune system (PMID: 24108437). The iNKT TCR is characterized by a conserved alpha chain (Vα24-Jα18 in humans) that recognizes these lipid-CD1d complexes with high affinity. Upon binding, the complex triggers the rapid release of a broad spectrum of cytokines, including IFN-gamma and IL-4, which can polarize the immune response toward either a pro-inflammatory or anti-inflammatory state (PMID: 17290283). This unique ability to bridge innate and adaptive immunity makes the complex a target for synthetic agonists like alpha-galactosylceramide (KRN7000) in treating cancer, infections, and autoimmune disorders (PMID: 29461216). Therapeutic challenges include the potential for iNKT cell anergy following overstimulation and the risk of systemic cytokine-related toxicities (PMID: 22343561).
