Antigen-presenting glycoprotein CD1d is a non-polymorphic, MHC class I-like molecule that specializes in the presentation of lipid and glycolipid antigens to Natural Killer T (NKT) cells (UniProt: P15813). The CD1d–lipid antigen complex is specifically recognized by the semi-invariant T-cell receptor (TCR) of invariant NKT (iNKT) cells, triggering a rapid and potent immune response characterized by the secretion of both Th1 and Th2 cytokines (PubMed: 17291281). This interaction serves as a bridge between the innate and adaptive immune systems, playing a critical role in tumor surveillance, defense against pathogens, and the regulation of autoimmune responses (PubMed: 21368833). Therapeutic strategies often involve the use of synthetic glycolipid agonists, such as alpha-galactosylceramide (KRN7000), to stabilize the complex and modulate iNKT cell activity for treating cancer or infectious diseases (PubMed: 24108444). However, overstimulation can lead to cytokine-related toxicities or long-term iNKT cell anergy, presenting a challenge for clinical application (PubMed: 11861601).
