Overview

The antigen-presenting machinery (APM) of autologous dendritic cells (DCs) refers to the integrated cellular system responsible for processing and displaying antigens to the adaptive immune system. This machinery includes key components such as the proteasome for protein degradation, transporters associated with antigen processing (TAP) for peptide translocation, and major histocompatibility complex (MHC) class I and II molecules for surface display [1]. In therapeutic contexts, autologous DCs are extracted from a patient and manipulated ex vivo to load specific antigens into this machinery [2]. The goal is to present these antigens to T cells upon re-infusion, thereby overcoming the immune evasion strategies often employed by tumors, such as the downregulation of their own APM [3]. This approach leverages the unique status of dendritic cells as the most potent professional antigen-presenting cells to initiate a robust anti-tumor or anti-viral response [4]. While not a single molecular target, the APM serves as the functional platform for several cell-based immunotherapies, most notably Sipuleucel-T for prostate cancer [5].
