Overview

The process of antigen-specific activation of CD4-positive and CD8-positive T lymphocytes refers to the mechanism by which T lymphocytes recognize specific antigenic peptides presented on major histocompatibility complex (MHC) molecules by antigen-presenting cells (APCs). CD4-positive T cells (helper T cells) are activated by peptides presented on MHC class II molecules, leading to immune system coordination, including B cell help, cytokine production, and regulatory functions[2][3][5][7][8]. CD8-positive T cells (cytotoxic T lymphocytes) are activated by peptides presented on MHC class I molecules, enabling them to directly kill infected or malignant cells[2][7][8]. Effective activation of both T cell subtypes requires co-stimulatory signals, often provided by CD28-CD80/86 interaction, and helper signals from CD4-positive cells enhance CD8-mediated responses[5][6]. This antigen-specific process is central to adaptive immunity and underlies the efficacy and side effects of immunotherapies, vaccines, and immune-modulating drugs.
