Antigen-specific B-cell receptors (BCRs) recognizing non-typeable Haemophilus influenzae (NTHi) surface antigens are specialized transmembrane immunoglobulins that mediate the adaptive immune response to NTHi infections. NTHi is a major human pathogen responsible for a significant burden of respiratory diseases, including acute otitis media in children and exacerbations of chronic obstructive pulmonary disease (COPD) in adults [Murphy et al., 2009, Journal of Infectious Diseases]. These BCRs recognize and bind to specific conserved surface proteins such as Protein D, P6, and various outer membrane proteins (OMPs), which are essential for bacterial adhesion and colonization [Forsgren et al., 2008, Clinical Infectious Diseases]. Upon antigen binding, the BCR initiates intracellular signaling pathways that lead to B-cell proliferation, affinity maturation, and differentiation into antibody-secreting plasma cells. In drug development, these receptors are the primary focus of vaccine strategies aimed at inducing protective humoral immunity, and they serve as the template for developing therapeutic monoclonal antibodies to neutralize NTHi and enhance opsonophagocytosis [Pichichero, 2013, Vaccine]. The study of these BCRs is also crucial for understanding why certain populations, such as those with COPD, fail to mount an effective immune response despite repeated exposure to the bacteria [King, 2012, Clinical & Experimental Immunology].
Vaccines containing NTHi surface antigens bind to and activate these B-cell receptors, leading to the production of protective antibodies and memory B cells.
