Overview

AP-1 complex subunit gamma-1 (AP1G1) is a large subunit of the clathrin-associated adaptor protein complex 1 (AP-1), which is a heterotetrameric protein complex essential for the intracellular sorting and vesicular transport of membrane proteins, particularly between the trans-Golgi network (TGN), endosomes, and the plasma membrane. AP1G1 is critical for the formation and cargo selection of clathrin-coated vesicles in polarized and non-polarized cells. Its function is vital for development, as loss-of-function mutations are embryonically lethal in animal models. Mutations or altered expression of AP1G1 have been linked to neurodevelopmental disorders and have roles in breast cancer progression, reflecting its importance in endomembrane trafficking, cellular polarity, and signaling[1][2][3][4][5][7].
