Overview

Apical membrane antigen 1 (AMA1) is a critical type I transmembrane protein found in Plasmodium falciparum and other apicomplexan parasites [1.1.1]. It is primarily localized in the micronemes and later on the surface of merozoites, where it plays an essential role in the invasion of host erythrocytes by forming a "moving junction" through interaction with rhoptry neck proteins, specifically RON2 [1.1.4, 1.3.1]. AMA1 is also involved in the invasion of hepatocytes by sporozoites [1.2.1]. Due to its indispensable role in the parasite life cycle, it has been a primary target for blood-stage malaria vaccines [1.3.4, 1.4.4]. However, the protein's ectodomain is highly polymorphic, particularly in Domain I, which allows the parasite to evade host immune responses and presents a significant challenge for developing a vaccine that provides broad, strain-transcending protection [1.2.2, 1.2.4]. Therapeutic strategies include recombinant protein vaccines, viral vector-based vaccines, and inhibitory peptides designed to block the AMA1-RON2 interaction [1.1.4, 1.3.5, 1.4.2].
