Overview

APOA1 antisense RNA (APOA1-AS) is a long non-coding natural antisense transcript situated opposite the APOA1 gene in the apolipoprotein gene cluster, chromosome 11q23.3[9]. It negatively regulates APOA1 transcription—primarily in the liver, small intestine, and colon—by promoting suppressive epigenetic modifications that repress APOA1 mRNA synthesis. Dysregulation of APOA1-AS is implicated in decreased HDL production and increased risk of cardiovascular and metabolic diseases. Experimental modulation of APOA1-AS, such as with antisense oligonucleotides, is being investigated as a therapeutic strategy to upregulate protective APOA1 protein expression in relevant disease states[7][9][1].
