Overview

The Apolipoprotein B receptor (APOBR) is a macrophage cell-surface receptor that binds specifically to apolipoprotein B48, which is present in dietary triglyceride-rich lipoproteins such as chylomicrons and very low-density lipoproteins. Predominantly expressed in reticuloendothelial cells including monocytes, macrophages, and endothelial cells as well as adipose tissue, APOBR mediates the uptake and internalization of these lipoproteins. This process allows for the supply of essential lipids and lipid-soluble vitamins to these cell types. However, in conditions of elevated plasma triglycerides, excessive activity of this receptor leads to increased lipid uptake by macrophages, resulting in foam cell formation and thereby promoting atherosclerosis and endothelial dysfunction. Expression of APOBR is regulated, with nuclear receptor pathways such as those involving PPARα and PPARγ capable of downregulating its activity and thus protecting against excessive lipid accumulation and potential atherogenesis.
