Overview

APOE mRNA is the messenger RNA product of the human APOE gene (chromosome 19), and it codes for Apolipoprotein E, a protein critical for lipid transport and cholesterol metabolism in both the periphery and the central nervous system. Differential expression and splicing of APOE mRNA lead to isoforms implicated in neurodegenerative and cardiovascular diseases, with the E4 allele conferring significantly increased risk for late-onset Alzheimer's disease, in part due to altered brain cholesterol homeostasis and increased amyloid plaque deposition[1][2][5]. Therapeutic strategies targeting APOE mRNA typically aim to reduce aberrant Apolipoprotein E levels, modify isoform expression, or influence downstream lipid metabolism[2][5]. Note: While protein targets (Apolipoprotein E) are more commonly referenced, APOE mRNA is an increasingly important molecular target in modern RNA-based drug development.
