Additionally, Apolipoprotein(a) mRNA is targeted by drugs like pelacarsen and olpasiran to reduce Lipoprotein(a) levels, an independent risk factor for atherosclerosis. These therapies typically work by inducing sequence-specific degradation of the target mRNA in hepatocytes, often facilitated by GalNAc conjugation for liver-specific delivery. The use of mRNA-targeting strategies allows for the modulation of lipid pathways that were previously considered undruggable by traditional small molecules.
