Overview

Apoptosis facilitator Bcl-2-like protein 14 (BCL2L14) is a member of the BCL-2 protein family, involved in the regulation of cell death (apoptosis) and cellular homeostasis[2][3][1]. It is distinguished by its non-typical structure: several isoforms exist (produced by alternative splicing), with the long isoform containing BH2 and BH3 domains, and shorter isoforms containing only BH3[1][4]. The gene is highly expressed in testicular tissue and various organs of the gastrointestinal tract, where it may contribute to mucin network stability and epithelial barrier function[1][4]. Though originally studied as a pro-apoptotic factor (especially for its BH3-only isoform), recent research suggests BCL2L14 also plays important roles in immune modulation, cell trafficking, and is implicated in tumor suppression. Reduction or loss of BCL2L14 has been linked to increased susceptibility to tumorigenesis, particularly in the colon. Its dysfunction or altered expression is found in various human cancers. While it is not a current direct drug target in clinical use, it is recognized as a candidate for therapeutic intervention, and experimental studies have attempted to modulate its activity with BH3 mimetics and other agents[1][3][4].
