Apoptosis inhibition refers to the molecular processes and factors that prevent or delay programmed cell death (apoptosis). This is a critical regulatory mechanism in multicellular organisms, ensuring cell survival under normal physiological conditions but also contributing to disease when dysregulated. The inhibition of apoptosis is primarily mediated by specific proteins and signaling pathways that block the activation of caspases—the central executioners of apoptosis—or otherwise promote cellular survival. Key molecules include Inhibitor of Apoptosis Proteins (IAPs) and Anti-apoptotic Bcl-2 Family Members. Signaling Pathways Involved: PI3K/Akt; NF-κB; MAPK. Overexpression/mutation leading to excessive inhibition contributes significantly to cancer development/progression by allowing abnormal cells to evade death signals. Conversely, insufficient inhibition may result in degenerative diseases due to excessive cell loss. Therapeutic approaches aim either at restoring apoptotic sensitivity in cancer cells or protecting healthy cells from inappropriate death.
