Apoptosis Signal-Regulating Kinase 1 (ASK1), also known as MAP3K5, is a key component of the mitogen-activated protein kinase (MAPK) signaling pathway. It acts as an upstream activator of c-Jun N-terminal kinases (JNKs) and p38 MAPKs in response to various cellular stresses, including oxidative stress, endoplasmic reticulum stress, and calcium influx. Through these pathways, ASK1 regulates apoptosis, inflammation, and other stress responses. ASK1 activity is tightly regulated by multiple mechanisms involving proteins such as thioredoxin (Trx) and calcium/integrin-binding protein 1 (CIB1).
