Apoptosis stimulants are agents that promote or trigger apoptosis, the programmed cell death process. These stimulants act by activating key molecular pathways that lead to cellular self-destruction, particularly in cells that are damaged, infected, or malignant. They target the extrinsic (receptor-mediated) and intrinsic (mitochondrial) pathways, ultimately activating caspase cascades and leading to cell death. Common strategies include directly activating death receptors, mimicking pro-apoptotic BH3-only proteins, or inhibiting anti-apoptotic factors like BCL-2/BCL-xL. Alterations in apoptotic signaling contribute not only to tumor development but also resistance to therapy. Many anticancer drugs exploit this vulnerability by stimulating apoptosis selectively in cancerous tissues.
