Overview

Apoptosis-stimulating of p53 protein 1 (PPP1R13B, also known as ASPP1) is a regulatory protein that enhances the pro-apoptotic functions of p53-family proteins by promoting their DNA binding and transcriptional activation at the promoters of proapoptotic genes[1][2][3][5][8][9]. It contains ankyrin repeats and an SH3 domain involved in protein-protein interactions and acts as a molecular scaffold. ASPP1 is mainly expressed in the cytosol and nucleoplasm and plays a key role in the intrinsic apoptotic pathway mediated by p53. It is implicated in cancer progression, where its downregulation is linked to epithelial-mesenchymal transition, increased invasion, and metastasis, particularly via modulation of the NF-κB/Snail2 pathway[4][6]. PPP1R13B has also been associated with rare syndromes such as Noonan syndrome 3 and 7[2]. No drugs are presently known to interact directly with PPP1R13B, but its biomarker potential and regulatory function in apoptosis make it of interest in oncology and cell death research.
