Overview

Aquaporin-11 is an intracellular member of the aquaporin family with distinct sequence features, including a conserved C-terminal NPA motif and a unique N-terminal NPC (Asn-Pro-Cys) motif. Unlike classical plasma membrane aquaporins, AQP11 localizes primarily to the endoplasmic reticulum and possibly other intracellular membranes, where it facilitates water, glycerol, and hydrogen peroxide transport, contributing to organ homeostasis, protein folding, redox balance, and development. In the kidney, AQP11 is essential for proximal tubule function and postnatal renal development; mice lacking AQP11 rapidly develop fatal renal cysts and failure. Its channel activity helps maintain homeostasis under physiological and stress conditions, including ER oxidative stress responses. While its dysfunction is linked to specific renal diseases and skin disorders, AQP11 is not a current therapeutic target, and no drugs are available that directly modulate its activity.
